Found on Canada's Baffin Island, Nettilling Lake is the largest lake on an island.  It helps that Baffin Island is Canada's largest island.

Manitoulin Island
Located in Canada's Lake Huron, Manitoulin Island is the world's largest island in a lake.  At a little over 1,000 square miles, the island is actually a tiny bit larger than Luxembourg.  

Manitoulin itself contains lakes that contains islands. Most famously, it is the home to Treasure Island, which is the largest island on a lake on an island on a lake.  Manitoulin Island also houses Manitou Lake, the largest lake on an island in a lake.

Why Canada?
Canada has the odds stacked in its favor.  In terms of land mass, Canada is the second largest country in the world and; thanks to the retreating glaciers at the end of the last Ice Age, it is home to over half of all the world's lakes.  Depending on one's definition of lake, some believe Canada actually has 60% of the world's lakes.

Canada can go even further.  The nation also claims to possess the largest island in a lake on an island in a lake on an island. This NASA article provides a little more information about the unnamed island. Live Science also published an article on the island.
